Stenny's last-minute goal hero from the first match, Alex Williams, is available after a ban.
Scott Dalziel remains a doubt because of a knee injury, while goalkeeper Chris McCluskey is fit again.
Williams scored in the 90th minute as Stenhousemuir came back from 2-0 down to force a replay on 20 November.
Threave Rovers manager Dave McVittie: "We matched Stenhousemuir in all departments in the first game so we can be confident although the dynamics of the tie have changed with David's move and of course he will be missing from our line up.
"One thing remains the same in that it would be great to win through to face local rivals Stranraer in the next round."
Stenhousemuir manager David Irons: "I know I let people at Castle Douglas know what was happening throughout.
"Unfortunately the timing was not ideal and it is a bit awkward as it is an incomplete tie between the sides.
"Threave feel aggrieved that I know their team inside out but I am not playing in it so it will be up to the guys on the park to do the business."
